چکیده

Clinically relevant differences amongst therapeutic antibodies against the same target may relate to their unique molecular attributes. Differences in profile across domains of psoriatic disease between guselkumab (GUS) and risankizumab (RIS) have been observed. To explore potential mechanisms underpinning this, we studied GUS, a fully human IgG1 specific for interleukin (IL)-23 with native Fc region, RIS, humanized anti-IL-23 mutated region. We compared binding functional characteristics antigen-binding regions these antibodies. GUS RIS displayed comparable picomolar affinities IL-23 by KinExA surface plasmon resonance assays, equivalent potency (IC50=0.2 nM) inhibition IL-23-induced STAT-3 phosphorylation peripheral blood mononuclear cells. However, cells transfected individual Fcg receptors (FcgRs), showed strongest CD64 (FcgR1), while negligible any FcgRs, virtue its Furthermore, interferon (IFN)g-primed monocytes, labeled dose-dependent flow cytometry, did not. on monocytes not trigger activation as shown lack cytokine or chemokine production. Importantly, CD64-bound was able bind IL-23, detected anti-p40 staining. In conclusion, uniquely binds both CD64+ myeloid IL-23. phagocytes are enriched skin serve dominant source. Further studies warranted generate additional evidence supporting this hypothesis.

Cancer therapeutic antibodies come of age: targeting minimal residual disease.

Ten years after the first clinical application of Rituximab, an anti-CD20 recombinant monoclonal antibody, immunotherapy has become common practice in oncology wards. Thanks to the great diversity of the immune system and the powerful methodology of genetic engineering, the pharmacologic potential of antibody-based therapy is far from exhaustion. The recent application of Trastuzumab, an antibo...

Targeting MICA with therapeutic antibodies for the treatment of cancer

MICA and MICB, along with ULPBs, are ligands for the activating receptor NKG2D expressed on NK cells and subsets of T cells in Human. NKG2D ligands are induced by cellular stress and pathogen infections. Their expression is tightly regulated by complex mechanisms both at the mRNA and protein levels. In the case of MICA and MICB, more than 65 and 30 alleles respectively were described with diffe...

Therapeutic targeting in nanomedicine: the future lies in recombinant antibodies.

The unique chemical and functional properties of nanoparticles can be harnessed for the delivery of large quantities of various therapeutic biomolecules. Active targeting of nanoparticles by conjugating ligands that bind to target cells strongly facilitates accumulation, internalization into target cells and longer retention at the target site, with consequent enhanced therapeutic effects. Interleukin-17A and interleukin-23 in morphea

INTRODUCTION Morphea is a disease included in the group of scleroderma type autoimmune diseases. Interleukin (IL)-17A may play a role at every stage of its pathogenesis. The study aimed at evaluation of IL-17A and IL-23 (as the main cytokine which is supposed to stimulate and maintain synthesis of IL-17) in pathogenesis of morphea. MATERIAL AND METHODS The studies were performed on 41 blood s...

Therapeutic Applications of Monoclonal Antibodies in Multiple Sclerosis

Despite the various therapies available, the use of monoclonal antibodies is a highly specific approach that has only recently been of interest to researchers. The properties of antibodies have led to their use in the treatment of various diseases, including cancer, Alzheimer's disease, diabetes and multiple sclerosis (MS). MS, a chronic inflammatory disease, occurs commonly in young adults. Th...
